Tender Details

The NAKO should be genetically characterized in order to enable the processing of genetic questions. For this purpose, all participants in the basic examination should be genotyped with a genetic array that covers the genome sufficiently for imputation and is also disease-specificche SNPs that are suggested by the relevant expert groups.

Key information about the samples within the NAKO project

The NAKO Health Study (NAKO) is a long-term population study. A core component of the study is the collection of various biomaterialsprocessing, storage, analysis and distribution for scientific evaluation.

The collected biosamples (exclusively human biological material) are stored in cryo-tubes (CR) at -80 C (serum, buffy coat, urine, saliva, stool, genomic DNA) or at -180 C (serum, plasma, erythrocells, urine). The DNA was isolated from the buffy coats using Perkin Elmer magnetic beads technology on the chemagic Prime 8 system. The isolated DNA samples generally meet the standard quality criteria for purity and yield for a subsequent chip-based genotyping:

(1) DNA concentration and volume 20 ng/yl in 400 yl (99.8%),

(2) absorption ratio 260/280 nm 1.8 and (3) absorption ratio 260/230 nm 1 .5 (92.0%).
