Description : Protein toxicity can be due to various mechanisms of action at molecular level (e.g. ion channel inhibitor activity, metal ion binding, etc), or be associated to biological processes (e.g. hemolysis, lipid catabolic process, defence response) and/or on the basis of the cellular component affected (Palazzolo et al. 2020). Proteins under assessment can be tested using in vitro systems, this efficiently integrating in silico predictions. The target of this work is to gather information on the available in vitro systems used to test the above toxicity mechanisms, processes or relevant cellular components and to evaluate their applicability to testing protein toxicity. Following the identification of in vitro tools, testing strategies for selected toxic proteins (i.e. identification of fit-for purpose battery of in vitro tools for a selection of toxic proteins) should also be developed.
