Obtaining a Modular Confocal STED Microscope with Pulsed 1 and 2 Photon Excitation and 4 STED Lasers.

The Confocal STED Microscope should be suitable in its basic configuration for multi-user operation and offer an experienced user the opportunity, after a short EinarThe STED microscope is to be modularly adaptable to the specific measurement conditions by means of exchangeable components. The requirements beyond the standard applications are:
Simultaneous 6-channel recording with pulsed excitation and STED:
The STED microscope is designed to measure the conformational dynamics of individual enzymes in vitro and in lecells using picosecond time-resolved Foerster resonance energy transfers (smFRET), homo-FRET and time-resolved fluorescence anisotropy. The full bandwidth of picosecond pulsed excitation with laser wavelengths from 440 nm to 640 nm for single-photon excitation (1P) must be included. The highest sensitive single-photon detection should be performed simultaneously in6 channels (for 3 spectral regions and each separated by anisotropy) take place. Preferred detectors are time-optimized APDs with the lowest possible dark counting rate (less than 100 counts / s). Ideally, in each case 2 of the pulsed lasers can be switched in freely configurable alternating excitation sequences in the nanosecond range (nsALEX or PIE),
3 pieces of pulsed STED laser adaptive beamforming:
The STED microscope is designed to excite fluorescent proteins (mNeonGreen or mScarlett) as well as the entire range of STED-suitable organic fluorophores with emission maxima from 500 nm to 700 nm with picosecond pulsed STED lasers at 590 nm and 775 nm. A third pulsed 660 nm STED laser is required for easy retrofitting. A programmable Spatial Light modulator (SLM) to enable the adjustment of the shape of the focus for 2D and 3D STED,
- multi-photon excitation for STED with 4-channel detection:
A tunable fs titanium sapphire laser system (TiSa) for two-photon excitation between 800 nm 1 300 nm must be coupled in and synchronized with the 3 pulsed STED lasers. 4 highly sensitive GaAsP photomultipliers are to be useddetect fluorescence lifetime and time resolved anisotropy measurements in two spectral ranges. These need not be read out simultaneously with the 6 APD detectors. The accompanying 4-channel TCSPC electronics must consist of synchronized modules with sub-picosecond time resolution,
- Extra microscope port for free-jet coupling 1 ultra-narrow band STED laser:
The STEDMicroscope must be modular designed so that the user can implement new STED methods on a suitable port of the microscope stand. In particular, a STED de-excitation with an ultra-fine-band laser system at 589 nm is required, which can be tuned exactly to the absorption line of a sodium vapor-filled gas cuvette. In addition, a multi-channel GaAsP PMT with TCSPC Erequired for the combination of fiber-based image scanning microscopy with STED in the z-dimension (ISM-STED).

Worth the awarding authorityIt is not an inadmissible double application if subcontractors are involved by different bidders. However, it is imperative that the subcontractor is not aware of the offer prices of the provider.
